Inserting a substrate into an enzyme will often cause the enzyme to change its shape.  This can cause a tension on the substrate at a designated bond allowing the larger molecule to be broken into two or more pieces.  These no longer fit the enzyme and are released.  This resets the enzyme for the next substrate.  Shape changes of the enzyme may bring side chains of the active site closer to portions of the substrate an put forces that help to break the bonds.
